=== Alzheimer's Disease === In contrary, big dynorphin exhibits neuroprotective properties against amyloid-β (Aβ) accumulation in Alzheimer's disease. It reduces the hydrophobicity and slows the aggregation kinetics of Aβ40, the most abundant amyloid species, resulting in around 2-fold reduction in amyloid aggregation compared to Aβ alone. In human neuroblastoma cell cultures, big dynorphin pre-incubated with Aβ40 significantly increased cell viability to compared to Aβ40 alone. A bowel obstruction is generally a mechanical obstruction of the gastrointestinal tract and can occur anywhere from the Ligament of Treitz to the anus. When the obstruction affects only the small intestine, it is generally referred to as a small bowel obstruction to distinguish it from a colonic obstruction, which may or may not affect the small intestine. The distinction helps to narrow the possible causes and treatment. Common causes of small bowel obstruction include post-operative adhesions, hernias, intussusception, and intraabdominal tumors. Common causes of colonic obstruction include primary colon cancer, volvulus and post-operative adhesions. When the ileocecal valve is competent, colonic obstruction may manifest as gaseous distention of the colon, but not the small intestine; when the ileocecal valve is incompetent, it does not prevent retrograde passage of air and stool and a colonic obstruction will cause dilation of both large and small bowel. Bowel obstructions can be partial or complete. They can be differentiated on imaging by the intestinal gas pattern. Partial obstructions will have gas distal to the obstruction, whereas a complete obstruction will not. Sounds of "rushes and tinkles" are associated with partial obstructions and represent brief passages of fluid and gas (respectively) through the partial obstruction. Complete obstructions do not make these sounds.
ALD is caused by mutations in ABCD1, located at Xq28 and demonstrates X-linked recessive inheritance. The gene ABCD1 encodes a peroxisomal membrane transporter which is responsible for transporting very long chain fatty acid substrate into the peroxisomes for degradation. Mutations in this gene that interfere with this process cause this syndrome. Males with an ABCD1 mutation are hemizygous, as they only have a single X chromosome. Female carriers will typically avoid the most severe manifestations of the disease, but often become symptomatic later in life. Although the detection of an ABCD1 mutation identifies an individual who is affected with a form of ALD, there is no genotype–phenotype correlation. Within a family, there will often be several different phenotypes, despite the presence of the same causative mutation. In one case, a family with six affected members displayed five different phenotypes. There are no common mutations that cause ALD, most are private or familial. Almost 600 different mutations have been identified, approximately half are missense mutations, one quarter are frameshifts, with in-frame deletions and splicing defects making up the remainder. === Airspace restrictions === At the direction of the US Department of Defense, the US Federal Aviation Administration (FAA) banned flights in an area around Ceiba, Puerto Rico, for "special security reasons" from 1 November 2025 to 31 March 2026. On 21 November, the FAA urged airlines flying over Venezuela at any altitude to exercise caution due to the "worsening security situation and heightened military activity in or around Venezuela". Increased GPS interference, linked to military action by both the US and Venezuela, made flight dangerous; by 25 November, at least eleven airlines had temporarily suspended flights to Venezuela. After threatening to permanently suspend the clearances of airlines that did not resume flights within 48 hours, on 26 November, Venezuela revoked the licenses of six airlines they said had "joined actions of state terrorism promoted by the United States". While Venezuela has authority over its airspace, on 29 November, Trump announced that airspace over the country should be considered entirely closed, days after he stated that the US would begin to strike land targets "very soon". The Washington Post stated that "such a move is sometimes a first step ahead of airstrikes"; while Trump cannot legally close Venezuela's airspace and he did not impose a no-fly zone, the "threat ... might be enough to prompt commercial airlines to halt flights over Venezuela". Venezuelan officials condemned Trump's statement as a "colonialist threat", stating that Venezuela rejects "orders, threats, or interference from any foreign power".
